Powder Rheology Useful Tools To Optimize the Formulation

Oral solid is a common method for delivering pharmaceutical drugs. The most preferable solid dosage form is tablet and capsule. The tablet dosage form has more than 50 market shares in the pharmaceutical industry. Various approaches are utilised to manufacture the tablet. i.e. Wet Granulation, Dry granulation and direct compression. The majority of pharmaceutical industries are using wet granulation or dry granulation approaches to manufacture tablets to avoid the risk of content uniformity issues which are likely to be observed in the direct compression approach. Wet granulation and dry granulation optimization require multiple processing operation evaluations and controls. By evaluating the powder rheology and understanding excipient properties, a direct compression approach can be utilised with a high degree of assurance for content uniformity a critical attribute for pharmaceutical solid dosage form. Two different molecules have been studied using powder characterization and simulation study to understand the powder behaviour segregation tendency and selection of the most suitable excipients for direct compression of selected drug molecules. Development of inhouse tools to understand powder segregation tendency apart from measurement of powder flow characteristic to develop direct compression approach.
